In vivo methodologies to identify and functionally characterize proteins involved in Fe/S protein maturation

Description of tasks:

  1. Identification of new yeast and human components in Fe/S biogenesis using, e.g., proteomics, yeast 2-hybrid and genetics
  2. Cellular localization of the identified proteins in yeast and human cells
  3. Gene knock-out of corresponding proteins in yeast and human cells. Phenotype analysis
  4. In vivo radiolabeling assay to measure the effect of gene-knocks on the incorporation of 55Fe into newly synthesized Fe/S proteins.
  5. Development of cell culture and animal models to study tissue-specific aspects of Fe/S related disorders
